Florian Sotoca opened the scoring late in the first half at the rain-soaked Stade Bollaert-Delelis, before Mathieu Udol doubled Lens’ lead with a header in the 76th minute.
Udol was then sent off for a reckless challenge on young substitute Khalis Merah, who has Algerian roots. Merah pulled one back for Lyon in the 85th minute, just seven minutes after coming on.
The victory was Yannick Cahuzac’s first as Lens coach since taking over from German manager Dino Toppmöller, who was dismissed last month after just six matches in charge.
The result ended Lens’ four-match winless run in the league and lifted them to seventh place, providing a major boost ahead of Tuesday’s Champions League home match against Portuguese side Sporting.
Lyon, meanwhile, remain two points behind league leaders Monaco.
Portuguese coach Paulo Fonseca’s side will host English club Crystal Palace on Thursday in the Europa League. Palace won the UEFA Conference League last season.
